血浆RGS2蛋白与血管紧张素Ⅱ及其1型受体在子痫前期中的作用

摘要:
目的:探讨妊娠期血浆中G蛋白信号调节因子2(RGS2)、血管紧张素Ⅱ(AngⅡ)及其1型受体(AT1R)在子痫前期(PE)发病中的作用及对疾病严重程度的影响。方法:选择2016年1~9月在南方医科大学附属深圳妇幼保健院住院分娩的PE孕妇50例为PE组(合并FGR组17例,非FGR组33例)。选择同期分娩的健康孕妇40例为对照组。采用ELISA法检测各组孕妇血浆中RGS2蛋白、AngⅡ及AT1R水平,并比较PE组(合并FGR组、非FGR组)与对照组间3种指标的差异,运用Logistic回归分析3种指标与PE的发生及严重程度的相关性。结果:(1)PE组血浆中RGS2蛋白、AngⅡ、AT1R水平及收缩压、舒张压均高于对照组,而取样孕周、分娩孕周及新生儿出生体质量均低于对照组,差异有统计学意义(P<0.05);合并FGR组、非FGR组间的RGS2蛋白水平、收缩压、舒张压差异无统计学意义(P>0.05),但均分别高于对照组(P<0.05);AngⅡ、AT1R水平及新生儿出生体质量在对照组、非FGR组及合并FGR组中两两比较,差异均有统计学意义(P<0.05)。(2)Logistic回归分析示RGS2、AngⅡ及AT1R是PE发生的独立危险因素[(粗OR>1,P<0.05;调整抽样孕周后的OR1>1,P<0.05;调整另外两种指标后的OR2值>1,P<0.05(除外AngⅡ)]。(3)RGS2蛋白、AngⅡ、AT1R水平与PE、收缩压、舒张压均呈正相关(P<0.05),AngⅡ、AT1R水平与新生儿出生体质量呈负相关(P<0.05),RGS2与新生儿出生体质量无相关性(P>0.05)。结论:血浆中RGS2蛋白、AngⅡ及AT1R可能与PE的发病机制相关,AngⅡ及AT1R可能与疾病的严重程度相关。

Abstract:
Objective: To investigate the role of serum RGS2 protein,AngⅡ and AT1 R in the Pathogenesis of Preeclampsia and it’s impact on the severity of the disease.Methods: Totally 50 patients with PE collected from Shenzhen Maternity and Child Healthcare Hospital,Southern Medical University during October 2015-September 2016 were recruited as PE group( PE with FGR group 17 cases and PE without FGR group 33 cases),40 cases of healthy pregnant women were collected as the control group.ELISA was used to detect the serum RGS2 protein,AngⅡand AT1 R levels.he differences between the groups were compared.Logistic regression was used to analyze the correlation between the three indexes and the severity of PE.Results:(1)In PE group plasma RGS2 protein,AngⅡ,AT1 R levels and systolic blood pressure,diastolic blood pressure,sampling gestational age,maternal gestational age,neonatal birth weight were higher than those in control group( P <0. 05); There was no significant difference on RGS2 protein level,systolic blood pressure and diastolic blood pressure between PE with FGR group and PE without FGR group( P>0. 05),There was significant difference on the AngⅡ,AT1 R level and newborn weight( P<0. 05).(2)Logistic regression analysis showed that RGS2、AngⅡand AT1 R were the independent risk factors of PE.( the crude odds ratio of RGS2,AngⅡ and AT1 R were > 1,P < 0. 05. After adjust the sampling of gestational age the OR values were > 1,P <0. 05.After adjust the other two indicators the OR values were >1,P < 0. 05,except for AngⅡ.).(3) RGS2 protein,AngⅡ,AT1 R levels were positively correlated with PE,systolic and diastolic blood pressure( P <0. 05). AngⅡ,AT1 R levels were negatively correlated with neonatal birth weight( P<0. 05).RGS2 was not related with neonatal birth weight( P>0. 05).Conclusions: Plasma RGS2 protein,Ang Ⅱ and AT1 R may be associated with the pathogenesis of PE and AngⅡand AT1 R may be associated with the severity of the disease.
