靶向阻断CXCR4对卵巢上皮性癌上皮间质转化影响的研究

摘要:
目的:探讨靶向阻断趋化因子受体4(CXCR4)对卵巢上皮性癌上皮间质转化(EMT)形成及肿瘤侵袭转移的影响。方法:CXCR4拮抗剂AMD3100作用后,采用MTT法、细胞克隆形成能力实验及Transwell侵袭实验分别检测卵巢癌细胞的增殖能力、细胞克隆形成能力及侵袭能力;采用Western blot法检测卵巢癌OVCAR3细胞中CXCR4以及EMT相关蛋白的表达。结果:AMD3100作用后,卵巢癌细胞OVCAR3的增殖能力、细胞克隆形成能力及侵袭能力明显受到抑制,且卵巢癌细胞中随着CXCR4表达的下降,EMT相关蛋白E-cadherin表达升高,而Vimentin、Snail表达则明显下降,且呈浓度依赖性,与空白对照组相比差异均有统计学意义(P<0.05)。结论:CXCR4可能调节EMT相关蛋白的表达,从而抑制卵巢癌的增殖、侵袭及转移能力。CXCR4有望成为治疗卵巢上皮性癌的新靶点。

Effect of Targeted Blockage of CXCR4 on the Epithelial Mesenchymal Transition in Epithelial Ovarian Cancer

Abstract:
Objective:To investigate the effect of targeted blockade of chemokine receptor 4(CXCR4) on epithelial mesenchymal transition and tumor invasion and metastasis in epithelial ovarian cancer.Methods:After the action of CXCR4 antagonist AMD3100,we used MTT,Cell cloning ability test and Transwell invasive experiment to detect the proliferation ability,cell clone formation ability and invasive ability of ovarian cancer cell;Western blot to detect the expression of CXCR4 and EMT related protein in ovarian cancer cells OVCAR3.Results:After the action of AMD3100,the proliferation ability,cell clone forming ability and invasion ability of OVCAR3 were inhibited obviously.The expression of EMT related protein E-cadherin increased with the decrease of CXCR4 expression,while Vimentin and Snail expression decreased significantly in a dose-dependent manner compared with the control group(P<0.05).Conclusions:CXCR4 may promote the proliferation of ovarian cancer as well as the invasion and metastasis process by regulating the expression of EMT related protein.CXCR4 may be a new target for the treatment of ovarian cancer.
